TBE is white to off-white instant granules, each bag of TBE instant granules can be prepared with 1 L of 1× TBE buffer, which is easy to operate and use. The main components of TBE are Tris, boric acid and EDTA. 1×TBE buffer contains Tris at a concentration of 89 mM, boric acid at a concentration of 89 mM, and EDTA at a concentration of 2 mM.

TBE is a nucleic acid electrophoresis buffer commonly used in biology, mainly used for DNA agarose gel electrophoresis. TBE has a strong buffer capacity, and it has better separation effect when it is used for electrophoresis of fragments smaller than 1 kb , and is also suitable for long-term electrophoresis. TBE is prone to hyperelectroosmosis when used in agarose gels, and the recovery of DNA fragments is reduced due to the interaction with agarose to form non-covalently bound tetrahydroxyborate complexes , so it is not recommended for DNA Fragment recovery .

Instructions 

1. Measure about 600 ml of distilled water into the beaker and place a magnetic stir bar in the beaker. 

2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of TBE instant granules, and stir the solution until completely dissolved. 

3. Add distilled water to the TBE solution in step 2, and make up to 1 L, which is 1×.

Note: The concentration of TBE working solution usually used is 0.5×. In this case, 1× working solution can be diluted before use.
